Correlation between Warm Reactive Autoantibodies and the Monocyte Monolayer Assay

ثبت نشده
چکیده

BACKGROUND: The monocyte monolayer assay (MMA) measures the adherence and phagocytosis of sensitized red cells with peripheral monocytes. The result of MMA may reflect the in vivo destruction of sensitized red blood cells (RBCs). STUDY DESIGN AND METHODS: Blood samples from 20 patients with warm reactive autoantibodies were investigated using the MMA. The Monocyte Index (MI) values > 5% were considered positive while MI values ≤ 5% were considered negative. RESULTS: Of the 20 patients, only one patient had a positive MMA result with an MI of 30%. All other 19 patients demonstrated a negative MI value (<5%). There was no correlation between serologically detectable warm reactive autoantibodies and the MMA or DAT. CONCLUSION: The final conclusion demands more comparative studies between the clinical and serologic features of patients with warm autoantibodies and the MMA tests. There are many variables that exist within the MMA which may have an effect on the outcome.

Can we consider erythrocyte sedimentation rate and C-reactive protein as a severity index in pemphigus vulgaris?

Background: Erythrocyte sedimentation rate (ESR) and C-reactive protein (CRP) are 2 inflammatory indicators that increase in many pathologic and physiologic conditions. Pemphigus vulgaris (PV) is an autoimmune skin disease in which autoantibodies against desmogleins cause acantholysis. In this study we have measured ESR and CRP levels in patients with PV to evaluate the relationship between an ...

Use of a Monocyte Monolayer Assay to Evaluate Fcγ Receptor-mediated Phagocytosis

Although originally developed for predicting transfusion outcomes of serologically incompatible blood, the monocyte monolayer assay (MMA) is a highly versatile in vitro assay that can be modified to examine different aspects of antibody and Fcγ receptor (FcγR)-mediated phagocytosis in both research and clinical settings. The assay utilizes adherent monocytes from peripheral blood mononuclear ce...

Altered control of self-reactive IgG by autologous IgM in patients with warm autoimmune hemolytic anemia.

Warm autoimmune hemolytic anemia (WAIHA) is characterized by an accelerated clearance of red blood cells (RBCs) associated with the presence of anti-RBC immunoglobulin (Ig)G autoantibodies. In the present study, we analyzed the self-reactive IgG and IgM antibody repertoires of patients with WAIHA using a technique of quantitative immunoblotting on a panel of whole tissue extracts as sources of ...

Development of an ELISA Based Method for the Detection of Pemphigus vulgaris Autoantibodies

For the evaluation of autoantibodies in Pemphigus vulgaris (PV), an indirect ELISA assay was developed by using a semi-purified human epidermal skin extracted with approximately a molecular weight (M.W.) of 130-160 kDa. The evaluation of Pemphigus IgG, IgM and IgA autoantibodies in 75 patients and 50 control sera indicated that this indirect ELISA assay was a useful method for the detection of ...
